目的探讨内嗅皮层注射Orexin—A对Sprague-dawley(SD)大鼠睡眠.觉醒周期以及对空间记忆的影响。方法选用健康成年雄性sD大鼠45只,体质量250—300g,采用同步记录大鼠脑电和肌电活动,观察Orexin—A及Orexinl受体阻断剂SB一334867内侧内嗅皮层微注射后大鼠睡眠-觉醒周期变化。采用T一迷宫交替选择次数,观察SB-334867内侧内嗅皮层微注射后大鼠T.迷宫选择正确率的变化。结果以人工脑脊液(ACSF)、二甲亚砜(DMSO)自身对照相比,分别给予大鼠内侧内嗅皮层双侧微注射OrexinA及SB一3348670.5p.1后3h(11:00—12:00、12:00~13:00、13:00—14:00)觉醒周期时间无影响(P〉0.05),给药后24h觉醒周期时间也无显著差异(P〉0.05)。微量注射药物后,Orexin.A(0.1nmoI/0.5txl)及SB-733467(6mmoI/0.5“1)在11:00—14:003h内对觉醒时间、NREM时间及REM时间均无影响(P〉0.05),即对3h内(1l:00—14:00)大鼠总睡眠觉醒时间无显著性差异(P〉0.05)。与DMSO组比较大鼠内侧内嗅皮层双侧微注射SB一3348670.5Ixl后,T一迷宫交替选择正确次数明显减少,正确率下降,有显著性差异(P〈0.001)。结论内侧内嗅皮层Orexin系统的支配可能参与了空间记忆,而不参与睡眠一觉醒周期调节。
Objective To study the effect of Orexin-A injected into medial entorhinal cortex (MEC) of rats on sleep-wake cycle and spatial memory of SD rats. Methods Forty-five healthy adult male SD rats, weighing 250 to 300 g, were included in this study. Their cerebral and myoelectrical activities were synchronically recorded, their sleep-wake cycle and accuracy rate of alternative selection in T-maze were observed after injection of Orexin-A and SB-334867. Results Oexin-A and SB-334867 showed no significant effect on sleepwake cycle, NREM and REM sleep time of rats 3 h after they were injected into MEC of rats compared with that of ACSF and DMSO (P 〉 0.05 ). However, the accuracy rate of alternative selection in T-maze was significantly lower after injection of SB-733467 than after injection of DMSO(P 〈0. 001 ). Conclusion The Orexin system in MEC may be involved in spatial memory but not in sleep-wake cycle.
