目的:采用细胞划痕实验,研究外源性TGF-β1(transforming growth factor beta)细胞因子对体外分离培养的原代癌相关成纤维细胞(CAFs)迁移特性的影响,探讨TGF-β1细胞因子对CAFs生物学性能的影响。方法:以口腔正常成纤维细胞(NFs)和未经处理的CAFs细胞为对照组,采用细胞划痕实验观察一定浓度的TGF-β1在不同时间作用后对CAFs迁移特性的影响。结果:与NFs组和未处理组CAFs相比,10μg/L的TGF-β1在不同时间点能够明显促进CAFs的迁移。结论:TGF-β1细胞因子能够明显促进CAFs的迁移,对CAFs的生物学性能具有重要影响。
Objective: To observe the effect of TGF-β1 on the migration of oral carcinoma--associated fibroblasts (CAFs), and to elucidate the role of TGF-β1 in the cellular characteristics of CAFs. Methods: Untreated CAFs and normal fibroblasts were set as the control group, wound healing assay was used to detect the migration of CAFs after treatment with TGF-β1 for varying periods. Results: Compared with the control groups, 10ng/ml TGF-β1 promoted the migration of CAFs. Conclusion: TGF-β1 can promote the migration of CAFs, and play a key role in CAFs biological characteristics.
