The polypyridyl ruthenium(Ⅱ) complexes of [Ru(bpy)2DMNP]^2+ and [Ru(bpy)2BOPIP]^2+ have been synthesized and their structures have been ascertained with elemental analysis and nuclear magnetic resonance spectra. The characteristics of the interaction of the complexes with CT-DNA were examined by Ultra-Vis absorption spectra, fluorescent spectra, viscosity measurements and gel electrophoresis technique. All data indicate that both complexes bind to double-stranded CT-DNA via the intercalative mode and [Ru(bpy)2DMNP]^2+ binds more avidly to CT-DNA than [Ru(bpy)2BOPIP]^2+. Noticeably, upon irradiation at 365 nm, the two complexes have been found to efficiently photocleave plasmid DNA, and the active oxygen species for the DNA cleavage in these complexes is singlet molecular oxygen.
