目的观察芪丹通脉片对犬慢性心肌缺血的预防作用,探讨其作用机制。方法将20只犬随机分为正常对照组,模型组,芪丹通脉片低、高剂量组,以注射异丙肾上腺素诱导心肌缺血模型;观察并比较各组犬心电图ST段变化(AST),及心肌血管内皮生长因子(vascularendothelialgrowthfactor,VEGF)、碱性成纤维生长因子(basicfibroblastgrowthfactor,bFGF)的表达水平。结果正常对照组未见ST段抬高及VEGF和bFGF表达;模型组可见ST段抬高及VEGF和bFGF表达;与模型组比较,芪丹通脉片低、高剂量组△ST值显著降低(P〈0.05),VEGF、bFGF表达水平显著升高(P〈0.05);与芪丹通脉片低剂量组比较,芪丹通脉片高剂量组△ST值有下降趋势,及VEGF、bFGF表达水平有升高趋势,但差异无显著性(P〉0.05)。结论芪丹通脉片能够预防慢性心肌缺血,促进心肌VEGF、bFGF表达可能是其作用机制。
Objective To observe effects of Qidan Tongmai Tablet (QDTMT), a traditional Chinese medicine prescription for invigorating qi to activate blood circulation,' in preventing chronic myocardial ischemia in dogs, and explore the mechanism. Methods Twenty dogs were randomly divided into normal control group, model group, low- and high-dosage QDTMT treated group. The dog model of chronic myocardial ischemia was induced by intramuscular injection of isoproterenol. The changes of ST segment (AST) of electrocardiogram (ECG), and the expression levels of vascular endothelial growth factor (VEGF) and basic frbroblast growth factor (bFGF) were observed and compared among four groups. Results ST segment elevation, and the expression of VEGF and bFGF were not found in normal control group; while they were found in model group. Compared with model group, the values of AST were significantly decreased in low- and high-dosage QDTMT treated group (P〈0.05), while the expression levels of VEGF and bFGF 0- were significantly increased (P〈0.05). Compared with low-dosage QDTMT treated group, the value of AST showed a decreasing tendency in high-dosage QDTMT treated group, while the expression levels of VEGF and bFGF showed an increasing tendency, but there was no significant difference between them. Conclusion QDTMT has preventive effects against chronic myocardial ischemia; the mechanism might be related with its promoting the expression of VEGF and bFGF in the cardiac muscle.
