摘要目的:探讨不同时长、不同剂量的贵州某品牌54°食用白酒对大鼠睾丸间质细胞内表皮生长因子(EGF)、嗜铬颗粒素A(CgA)的表达及血清睾酮水平的变化。方法:正常成年雄性SD大鼠,随机分为正常对照组及实验组。采集血清,取睾丸组织。采用免疫组织化学SABC法、图像分析、免疫印迹、化学发光法检测。结果:免疫组织化学显示,EGF、CgA阳性产物存在于睾丸间质细胞胞质内;不同时间点实验组各剂量组EGF及CgA免疫染色强度及平均光密度无明显改变。免疫印迹结果显示,实验组各剂量组EGF蛋白表达水平均无明显改变。各实验组的血清睾酮水平与正常对照组比较未见明显异常,差异无统计学意义。结论:在本实验设定的剂量和时长内,用该品牌54°食用白酒灌胃对睾丸间质细胞内EGF、CgA的表达及血清睾酮水平无明显影响。

英文摘要:

Objective: To discuss the effect of time and dose of some 54 degree liquor on the expression of epidermal growth factor (EGF) and chromogranin A(CgA) and the change of the serum testosterone level of rat Leydig ceils so as to provide basic data for scientific drinking. Methods: Normal adult male SD rats were randomly divided into normal control and experimental group. Serum and testicular tissue of normal control group and experimental group were collected for immunohistochemical SABC test, Lrnage analysis, Western blotting analysis and chemiluminescence study. Results: Immunohistochemical positive EGF and GgA protein were expressed in Leydig ceils within the cytoplasm. Compared with normal control group, EGF, CgA immuno staining intensity and the average optical density had no obvious change in each dose group. Western blotting showed no obvious change in the expression level of EGF in 4-week and 8-week, 12-week group of different doses. Compared with normal control group, serum testosterone level of experimental groups didn't present anything apparently unusual and there was no statistically significant difference. Conclusion : The synthesis and secretion of testosterone may not be affected by dose and time point of liquor in the experiment.
