目的通过实验观察复合干细胞因子的凝胶PluronicF-127对骨髓间充质干细胞(bone marrow mesenchymal stemcells,BMSCs)定向迁移的作用及其对大鼠下颌骨骨创伤后愈合的影响。方法①使用Transwell小室进行SCF凝胶组、空白对照组对骨髓间充质干细胞BMSCs的体外趋化实验,并对迁移细胞进行计数。②在30只清洁级SD大鼠双侧下颌骨分别制造直径3 mm骨缺损模型,统一在右侧注射复合材料,左侧为对照。并在术后1、2、4、6、8周分别取6只处死并取出下颌骨观察,拍摄X线片比较骨缺损情况;将标本固定切片后分别进行碱性磷酸酶与CD34免疫组化染色,进行染色评分和新生血管计数,t检验统计分析评判结果。结果体外培养的大鼠骨髓间充质干细胞,趋化实验结果显示SCF凝胶组的迁移细胞数量大于空白对照组(P〈0.05);下颌骨手术大鼠术后1~8周创口内复合材料逐渐吸收,术后碱性磷酸酶免疫组化染色评分实验组高于对照组(P〈0.05),术后1~2周实验组成血管数多于对照组(P〈0.05),而4~8周无明显区别(P〉0.05)。结论复合SCF的凝胶对BMSCs在体外有一定趋化作用,在大于自愈骨缺损的体内骨创愈合中能发挥促成骨和成血管作用,加快愈合进程。
Objective To detect the effect of stem cell factor(SCF) combined with PluronicF-127 gel on the directional migration of bone marrow mesenchymal stem cells(BMSCs),then investigate the effect on healing after repair of mandibular defects in rats.Methods ①Transwell chamber was used to observe the migration of BMSCs in SCF gel group and blank control group.Count the number of migrated cells.②Bone defects 3 mm in diameter were established in 30 clean SD rats' bilateral mandibles,on the right sides of which composite material was injected,while the left sides of which served as control group.6 rats were sacrificed separately in the 1st,2nd,4th,6th,8th weeks after surgery.The mandibles were taken out and the bilateral bone defects were observed by X-ray.The specimens were fixed,sliced and dealt with histochemical staining of alkaline phosphatase(ALP) and CD34 separately.The staining was scored.The growth of new vessels was counted.T test was used to analyze and judge the results.Results For bone marrow mesenchymal stem cells cultured in vitro,chemotaxis assay showed that number of migrated cells in SCF gel group was more than that in blank control group(P〈0.05).Composite material in the wound of rats was gradually absorbed 1-8 weeks after mandibular surgery.Scores of ALP immunohistochemical staining in SCF gel group was higher than that in blank control group after surgery(P〈0.05).Number of new vessels in experimental group was more than that in control group 1-2 weeks after surgery(P〈0.05),while two groups had no significant difference in number of new vessels during 4-8 weeks(P〈0.05).Conclusions Composite SCF-gel can migrate BMSCs to certain degree.Furthermore,it can promote new vessels' growth and new bones' formation and accelerate healing process of orthopaedic injuries in vivo which are larger than self-healing bone defects.
