目的探讨GSTA1基因在肝癌形成中的作用。方法应用RT-PCR方法检测35例AFB1诱发的树鼬肝癌、癌旁和癌前组织,以及35例人肝癌和癌组织中的GSTA1mRNA表达情况;应用免疫组化方法检测以上组织的GSTA1蛋白表达情况。结果树韵肝癌组织的GSTA1mRNA和蛋白表达水平均低于癌旁及癌前组织;人肝癌组织的GSTA1mRNA表达水平、蛋白阳性表达率和表达综合得分均显著低于癌旁组织(分别为P〈0.001、P〈0.05和P〈0.001)。结论GSTA1可能是肝癌发生发展的重要相关基因之一;在mRNA水平和蛋白质水平动态观察相关基因在肝癌形成过程中的表达变化有助于阐释肝癌发生的分子机制。
Objective In order to elucidate the role played by GSTA1 in hepatocarcinogenesis. Methods GSTA1 mRNA and protein expression levels were detected on HCC, para-HCC and pre-HCC tissues from 35 cases of tree shrews' HCC induced by Aflatoxin B1 , as well as on the HCC and para-HCC tissues from 35 cases of human HCC, through the methods of RT-PCR and immunohistochemical staining. Results The expression level of GSTA1 mRNA and protein in tree shrews' HCC tissues was lower than that in para-HCC and pre-HCC tissue. Its mRNA level in human HCC tissues was also significantly lower than that in para-HCC tissues (P〈0. 001). Both the positive expression rate and general score of GSTA1 protein in human HCC were significantly lower than that in para-HCC tissues (P〈0. 05 and P〈 0. 001, respectively). Conclusion GSTA1 might be one of the principal genes that play important roles in hepatocarcinogenesis. Dynamic observation of the target gene on its mRNA and protein expression level should be helpful in elucidating the molecular mechanism of hepatocarcinogenesis.
