肾阳虚证患者外周血差异表达microRNAs筛选及功能预测

目的探讨右归丸治疗肾阳虚证的免疫调控机制。方法筛选9例肾阳虚证患者,均给予右归丸口服,连服1个月。分别于治疗前后采集患者外周静脉血,制备外周血单个核细胞(PBMC),提取RNA,按照实时定量PCR(q PCR)实验流程,检测TLR4、My D88及NF-κBmRNA表达情况,并进行比较分析。结果治疗后,患者TLR4mRNA表达水平明显下调(P〈0.05),My D88、NF-κB mRNA表达水平均明显上调(P均〈0.05)。结论右归丸可以调节TLR4/My D88/NF-κB信号通路的表达,这可能是右归丸良性调节肾阳虚证免疫功能的分子机制之一。

英文摘要:

Objective It is to discuss the immune regulation mechanism of Yougui Pills on the Kidney-Yang deficiency syndrome. M ethods Nine Kidney-Yang deficiency patients were selected for one month treatment with Yougui Pills. Accumulated clinical scores of Kidney-Yang deficiency were assessed pre-treatment and post-treatment using a4 0- factor questionnaire. Nine patients were all observed with good therapeutic efficacy and were prepared for Realtime Quantitative PCR( q PCR) tests. 1 0 m L blood were collected to tract Peripheral Blood Mononuclear Cell( PBMC) for each patients pre-treatment and post-treatment with Yougui Pills. With the RNA from PBMC,we did q PCR tests to detect mRNA expressed value of three target genes of TLR4,My D88 and NF-κB. Results TLR4,My D88 and NF-κB genes of Kidney-Yang deficiency patients were significantly expressed after treated by Yougui Pills,the TLR4 gene was down regulated while the My D88 and NF- κB gene was up regulated. Conclusion Yougui Pills could regulate the expression of TLR4 / My D88 / NF-κB signal pathway,which may be one of the molecular mechanisms for Yougui Pills to regulate the immune function of Kidney-Yang deficiency syndrome.
