三七注射液对阿霉素肾纤维化大鼠ILK、MMP-7及肾小管上皮细胞转分化的影响

目的观察三七注射液对阿霉素肾纤维化大鼠肾组织整合素连接酶(ILK)、基质金属蛋白酶-7(MMP-7)、E-钙黏素(E-Cadherin)、α-平滑肌肌动蛋白(α-SMA)的影响。方法 120只SD大鼠随机分为5组:正常组(n=24)、假手术组(n=24)、模型组(n=24)、三七注射液组(n=24)和三七总皂苷组(n=24),12周后处死大鼠。观察肾功能、24 h尿蛋白定量、肾脏病理改变,免疫组织化学法和Western blotting观察大鼠肾组织ILK、MMP-7、E-Cadherin和α-SMA蛋白的表达。结果假手术组大鼠肾组织结构正常,模型组大鼠出现肾间质纤维化,三七注射液组和三七总皂苷组大鼠小管间质病变减轻。模型组大鼠血清BUN和Scr水平、24 h尿蛋白定量、肾小管间质病理评分增高,肾组织ILK、MMP-7、α-SMA蛋白表达上调,E-Cadherin下调,与假手术组相比较,差异均有统计学意义(P〈0.01);而三七注射液组和三七总皂苷组大鼠BUN和Scr水平、24 h尿蛋白定量、肾小管间质病理评分、肾组织ILK、MMP-7、α-SMA蛋白下降,E-Cadherin上调,与模型组相比较,差异均有统计学意义(P〈0.01或P〈0.05);三七注射液组和三七总皂苷组相比较,上述指标差异均无统计学意义(P〉0.05)。结论三七注射液的肾保护作用可能与下调ILK、MMP-7水平,抑制肾小管上皮细胞转分化有关。

英文摘要:

[Objective] To observe the effect of Sanqi injection on the protein expression in kidney of integrinlinked kinase(ILK), Matrix metalloproteinases- 7 (MMP- 7), alpha - smooth muscle actin (α- SMA) and E-Cadhefin in rats model with Adriamycin-induced kidney fibrosis. [Methods] 120 rats were divided into 5 groups randomly: normal group, sham group, model group, Sanqi injection group and Sanqizongzaogan group. The rats were sacrificed after 12 weeks. And the kidney function, albumen amount of 24 hours urine, kidney pathology were detected, and the protein expression of ILK, MMP-7, α- SMA and E-Cadherin in the kidney were detected by the way of immunohistoehemistry and western blotting. [Results] The kidney struc- tures both in the sham group and normal group are regular. And the renal interstitial fibrosis in the Model group rats is found. However, the kidney pathology is improved in the Sanqi injection group and Sanqizongza- ogan group. Our results indicated that, the level of Blood urea nitrogen and creatinine, Albumen amount of 24 hours urine, the semi-quantitative seores, the protein expression of ILK, MMP-7, α- SMA in the kidney in the Model group are higher than those in the sham group. However, the protein expression of E-Cadherin in the Model group is less than that those in the sham group. However, both Sanqi injection and Sanqizongzao- gan injection treatment could reduce index including the level of Blood urea nitrogen and creatinine, Albumen amount of 24 hours urine, the semi-quantitative scores, the protein expression of ILK, MMP- 7,α- SMA in the kidney, but up-regulating the protein expression of E-Cadherin. [Conclusion] Sanqi injection could pro- vide a nephroprotective role by down-regulating of the protein expression of ILK, MMP- 7, and inhabiting Epithelial-mesenchymal transformation.
