目的:探讨热休克蛋白90(HSP90)是否能够抑制肿瘤坏死因子α(TNF-α)和放线菌酮(CHX)协同诱导的细胞凋亡.方法:采用电穿孔技术建立稳定过表达HSP90的细胞克隆,应用激光共聚焦显微镜和流式细胞仪观察TNF-α/CHX协同诱导的细胞凋亡.结果:在稳定过表达HSP90的小鼠成纤维细胞系NIH3T3中,HSP 90能够抑制TNF-α/CHX协同诱导的细胞凋亡;在TNF-α/CHX协同诱导的细胞凋亡中,HSP 90持续稳定存在12 h以上.结论:HSP 90能够抑制TNF-α/CHX协同诱导的细胞凋亡.
Objective: To investigate whether heat shock protein (HSP90) could suppress the apoptosis synergistically induced by tumor necrosis factor-α (TNF-α) and cycloheximide (CHX). Methods: The cell clone with stable HSP90 overexpression was estabfished. The TNF-α/CHX induced apoptosis was detected by using laser confocal microscope and flow cytometry. Results: HSP90 suppressed the TNF-α/CHX induced apoptosis in NIH3T3 cells with stable HSP90 overexpression and existed stably for more than 12 hours. Conclusion: HSP90 could suppress TNF-α/CHX induced apoptosis.
