构建了一种针对肺癌标志物细胞角蛋白19的可溶性片段CYFRA21-1的三通道高精度光学检测系统。系统基于磁酶免疫分析实验的特点,通过巧妙的机械结构和精确的控制系统设计,利用FPGA高频计数功能、NIOS核控制功能,配合自己编写的上位机软件,实现了对CYFRA21—1的定量检测;在确保高精度的前提下,实现了3个通道的同时检测,大大节省了检测时间,提高了检测效率。通过多次实验验证,系统3个通道间干扰小、一致性好,相对偏差小于1%。在CYFRA21—1抗原溶液检测实验中,系统在CYFRA21—1浓度为2~100ng/mL范围内线性关系良好,相关系数为0.979。系统具有小型便携、灵敏度高、检测时间短和重复性好等特点,为疾病标志物的快速检测提供了强有力的工具。
A three-channel high-precision optical detecting system for lang cancer marker CYFRA21-1 is described. This system is based on the magnetic enzyme-linked immunoassay, depends on the skillful mechanical design and precise controlling system, uses FPGA to count the high-frequency pulse and NIOS to control the whole system,and achieves the quantitative detection of CYFRA21-1 with the help of my own PC software. On the premise of high precision,three channels can be used in the same time,which can increase the efficiency of detection significantly. After experimental verification, the interference among the three channels is little and the consistency is good, and the relative deviation is lower than 1 %. The CYFRA21 1 sample with concentration of 2-100 ng/mL was measured and the correlation coefficient between CYFRA21-1 concentrations is 0. 979. This system is small, has high sensitivity and short detecting time, so it could be a powerful tool for the rapid disease marker detection.
