目的探讨磷酸化真核细胞翻译起始因子4 E ( p- eukaryotic translation initiation factor 4 E, p-eIF4 E )和磷酸化MAPK作用激酶1( p- MAP kinase integrating kinase, p-Mnk 1 )在头颈鳞状细胞癌( head and neck squamous cell cancer, HNSCC)中的表达及其临床意义。方法HNSCC组织芯片结合免疫组化S.P法检测P-eIF4E和P—Mnkl在60例HNSCC和35例癌旁组织中的蛋白表达,分析两者表达与HNSCC临床病理特征的相关性。结果p-elF4E和P—Mnkl在HNSCC中的阳性表达率显著高于癌旁组织(P〈0.01),P-eIF4E的阳性表达率与HNSCC的分化程度、临床分期显著相关(P〈0.05);有淋巴结转移HNSCC的P.Mnkl阳性表达率显著高于无淋巴结转移者(P〈0.05)。HNSCC组织中P-eIF4E和P-Mnkl表达呈显著正相关(r=0.375,P=0.005)。结论HNSCC组织中P-eIF4E和P—Mnkl异常表达与HNSCC的临床进展和浸润转移相关,两者在肿瘤发生发展过程中有协同作用。
Objective To study the expression of p-eIF4 E and p-Mnk 1 in the head and neck squamous cell carcinoma (HNSCC) and its clinical significance. Methods Expression of p-eIF4E and p-Mnk 1 was detected in 60 samples of HNSCC and 35 of peri-cancer tissues by tissue microarray immunohistochemical S-P method. Results The positive expression rates of p-eIF4E and p-Mnkl in HNSCC were significantly higher than those of peri-cancer tissues (both P 〈 0.01 ). The expression of p-eIF4E was correlated with pathological grading and clinical stages of HNSCC ( P 〈 0.05 ). The expression rate of p-Mnkl in HNSCC with lymphatic metastasis was significantly higher than that without metastasis (P 〈 0.05 ). In addition, the expression of p-eIF4E was positively correlated with that of p-Mnk 1 in HNSCC ( r =0. 361 , P = 0. 005 ) . Conclusions The abnormal expression of p-eIF4 E and p-Mnk 1 is related to the development, invasion and metastasis of HNSCC. Co-expression of p-eIF4E and p-Mnkl has a possible synergistic action that may promote the carcinogenic process of HNSCC.
