针对抗肿瘤药物吉西他滨半衰期短,临床上必须使用大剂量持续静脉滴注和毒副作用较大的问题,制备了载吉西他滨原位温敏凝胶,考察了其体外释放行为及流变学性质,并对其荷4T1肿瘤模型小鼠瘤旁给药时的抗肿瘤药效和初步毒性进行了评价。结果表明:吉西他滨原位温敏凝胶能缓慢、匀速释放吉西他滨,与溶液剂相比,原位凝胶具有较好的抗肿瘤疗效和较低的全身毒性,为肿瘤的局部化疗提供了一个新思路。
Chemotherapeutic agent gemcitabine is generally used in clinic by continuous intravenous infusion due to its short half life,resulting in serious side effects.Gemcitabine-loaded thermosensitive hydrogel was prepared and evaluated with respect to both in vitro drug release and rheology and in vivo anti-tumor efficacy and systemic toxicity.The results show that gemcitabineloaded thermosensitive hydrogel can release drug at a slow and steady rate,improved in vivo tumor growth inhibition and alleviated systemic toxicity,indicating good potential of thermosensitive hydrogel for sustained-release administration of gemcitabine for local chemotherapy.
