对实验室的10株真菌进行筛选,得到一株具有高效木聚糖降解能力的菌株50647。通过ITS序列分析,并结合形态学鉴定确定其属于拟盘多毛孢属。该菌以燕麦秸秆粉和麸皮不溶性木聚糖作为C源,于25℃摇瓶光照培养,第3天木聚糖酶的酶活峰值为49.3U/mL,酶的比活力为1 066.2U/mg。经SDS-PAGE及酶谱分析,该菌株能够产生至少6种不同分子量大小的木聚糖酶组分。
In order to screen strains that have high efficient xylan degrading ability,liquid fermentation and culture,phylogenetic and morphological analysis,SDS-PAGE and zymogram analysis and other methods were used.Comparative analysis revealed that in 10 strains,50647 showed the best xylanase producing time and activity.After 3 days,the activity of xylanase in the culture supernatant reached to 49.3 U/mL,special activity was 1066.2U/mg;This strain was identified as Pestalotiopsis sp.Culture supernatant contained at least six xylanases with different molecular mass.
