目的:观察溃结灵含药血清对结肠上皮细胞损伤模型MUC-2及TGF-α蛋白含量的影响,进一步探讨渍结灵促进肠黏膜损伤修复的机理。方法:TNF-α(50ng/mL)干预大鼠结肠上皮细胞6h造成细胞损伤模型,采用Elisa方法检测不同浓度溃结灵含药血清对细胞损伤模型MUC-2及TGF-α蛋白含量的影响。结果:①结肠上皮细胞损伤模型组MUC-2蛋白含量较空白对照组升高(P〈0.05);②溃结灵含药血清高剂量组MUC-2蛋白含量较模型对照组升高(P〈0.05);溃结灵含药血清高剂量组TGF-α蛋白含量较模型对照组升高(P〈0.01)。结论:溃结灵含药血清可促进TNF-α致结肠上皮细胞损伤模型中MUC-2、TGF-α含量升高,进而促进受损肠黏膜上皮的修复。
Objective To establish the damage model of rat colonic epithelial cell, and observe the effect of serum containing Kuijieling on contents of MUC-2 and TGF-α in the cell damage model in order to explore the mechanism of Kuijieling promoting to repair the injury of mucous membrane. Methods The colonic epithelial cell was intervened with 50ng/mL TNF-α for 6h to induce the cell damage model. The contents of MUC-2 and TGF-α were detected by using Elisakit. Results (1)Compared with the blank control group, the MUC-2 content in the cell damage model group was obviously increased(P〈0.05).(2)The contents of MUC-2 and TGF-α in high- dosage group (10% Kuijielin drug-contained serum) were significantly higher than that in the model group (P〈0.05, P〈0.01). Conclusion Kuijieling drug-contained serum could boost the contents of MUC-2 and TNF-α in rat colonic epithelial cell damage model, which might promote to repair of the impaired colonic mucous membrane epithelium.
