目的研究7,3’-二甲氧基橙皮素(DMHP)对佐剂性关节炎(AA)大鼠免疫功能的影响。方法弗氏完全佐剂诱导大鼠AA模型,从造模第12天开始灌胃给予DMHP(22、44、88mg/kg),连续12d,MTT法检测刀豆蛋白A诱导的脾淋巴细胞增殖反应;酶联免疫吸附法测定脾淋巴细胞产生白介素-2(IL-2)的水平以及腹腔巨噬细胞(PM中)分泌的IL-1、IL-10、肿瘤坏死因子-a(TNF—a)水平;RT-PCR检测PMФ IL-1、IL-10、TNF-a mRNA的表达。结果DMHP(44、88mg/kg)能明显纠正AA大鼠低下的脾淋巴细胞增殖反应和脾细胞IL-2的产生;各剂量DMHP能不同程度地从基因和蛋白水平降低AA大鼠PM中产生过高的IL-1、TNF-a水平,同时上调AA大鼠PMФ产生过低的IL-10水平。结论DMHP体内给药能改善AA大鼠异常免疫功能,恢复细胞因子网络平衡,这可能是其治疗AA大鼠的机制之一。
Objective To investigate the effect of 7, 3'-dimethoxy hesperetin (DMHP) on immune function in rats with adjuvant arthritis (AA). Methods Freund's complete adjuvant was used to induce AA in rats. DMHP (22, 44, 88mg/kg) were administered intragastrically on 12th day after AA induction, once a day from day 12 to 21. Splenic lymphocyte proliferation response induced by concanavalin A was examined with MTT assay. IL-2 production of splenic lymphocytes and IL-1, IL-10, TNF-a productions of peritoneal macrophage (PMФ) were all determined by enzyme linked immunosorbent assay (ELISA). In addition, the levels of TNF-a, IL-1, IL-10 mRNA were determined by using RT-PCR. Results The suppressed lymphocyte proliferation and IL-2 production of splenic lymphocytes in AA rats were reversed by treatment with DMHP (44, 88 mg/kg). Meanwhile, DMHP could down-regulate IL-1, TNF-a productions of PMФ and up-regulate IL-10 production of PMФ at both protein and transcription levels in different degrees. Conclusion One possible mechanism of the therapeutic effect of DMHP on AA rat may be related to adjusting abnormal immune function in AA rats and keeping the balance of cytokine network.
