目的探讨参麦注射液对脑出血大鼠血肿周围区缺氧诱导因子1-α(HIF1-α)表达的影响及其意义。方法将50只SD大鼠随机分为正常组、假手术组、模型组、参麦组,模型组和参麦组组内又分为1d,3d,5d,7d共4个时间点,采用立体定位仪定位注射胶原酶Ⅶ造大鼠脑出血模型,观察大鼠脑出血后神经病理体征改变,免疫组织化学法检测血肿周围区HIF1-α的表达,以及参麦注射液治疗后的影响。结果与模型组比较,参麦组大鼠神经病理体征减轻明显(P〈0.01);正常组和假手术组脑组织神经细胞无HIF1-α表达,而模型组血肿周围区HIF1-α的表达1d时增多,3d时达到高峰,5d时表达下降,至7d时仍有少量表达,与模型组比较,参麦组各时间点的HIF1-α表达要少(P〈0.01)。结论参麦注射液可能通过促进血肿吸收、抑制血肿周围区HIF1-α表达而发挥神经保护作用。
Objective To study The effect of Shenmai Injection on the expression of hypoxia- inducible factor 1 - α (hypoxia inducible factor 1 - α, HIF1 - α) in the perihematomal area in rat with intracerebral hemorrhage and its significance. Methods Fifty rats were randomized into 4 groups:Normal group, sham operation group, model group and treatment group with Shenmai Injection, The rats in the model groups and treatment groups were prepared at 1 d, 3 d, 5 d and 7 d, respectively after operation. The models were established by injecting collagenase type Ⅶ stereotaxically. The pathologic signs of the nerve system were observed. The expression of hypoxia - inducible factor 1 - α in the perihematomal area were analyzed by irnmunohistochem. Results Compared with model groups, the pathologic signs of the nerve system were alleviated, hematoma were absorbed well and the structure of neurons were relatively integrated(P〈 0.01 ). There was no expression of HIF1 - α in the normal groups and sham operation groups. However, the expressions of HIF1 - α in the of rats neurons in the perihematomal area increased at 1st day, got to a peak at 3rd day, decreased gradually at 5th day and kept higher at 7th day. There were obvious significant differences between treatment group with Shenmai Injection and model groups(P 〈 0.01 ). Conclusion Shenmai Injection might have a protective effect on the rats after ICH, which might be activated by promoting the absorption of hematoma and inhibiting of the expression of HIF1 - α.
