M蛋白是新城疫病毒(NDV)的一个多功能结构蛋白。为了制备针对M蛋白的多克隆抗体,本研究通过克隆M蛋白部分基因片段至原核表达载体,重组蛋白在大肠杆菌中表达后进行了纯化。免疫新西兰大白兔获得血清并纯化后得到抗M蛋白多克隆抗体。Western-blot分析结果显示,该抗体具有良好的特异性。本试验成功制备的抗M蛋白多克隆抗体,为进一步研究M蛋白的生物学功能奠定了良好的基础。
Matrix protein of Newcastle disease virus is a multifunctional protein. In order to prepare its polyclonal antibodies against matrix protein,we cloned a fragment of matrix gene into prokaryotic expression vector,and the recombinant protein was expressed in Escherichfa colf and purified. New Zealand white rabbit was immunized with recombinant protein to obtain anti-M serum. The polyclonal antibodies was purified. Western-blot analysis showed that the polyclonal antibodies had good specificity against matrix protein. The polyclonal antibodies is useful for further research on biological functions of matrix protein.
