为提取高浓度的黄曲霉毒素B1,以大米为培养基培养黄曲霉,分别在不同温度梯度(15℃、20℃、25℃、30℃、35℃)和湿度梯度(0%、80%、85%、90%和95%和100%)下培养,每天测量一次菌斑直径的变化(共测18次),然后采用Logistic(一级)和Ratkowsky(二级)模型对黄曲霉菌斑大小进行拟合和检验,最后借助响应面模型对黄曲霉产毒条件进行优化.结果显示,在35℃和100%湿度的培养条件下,只需培养16~17d即可获得直径较大的黄曲霉菌斑,提取黄曲霉毒素B1浓度为3312ng/mL,单位面积黄曲霉毒素B1产量是优化前(1101.471ng/mL)的3倍.
Optimization of culture conditions for Aspergillus f lavus was carried out to extract high concentration of aflatoxin B 1.A.flavus was cultivated in the rice medium under different temperature gradients (15 ℃,20 ℃,25 ℃,30 ℃,and 35 ℃) and different humidity gradients (0%,80%,85%,90%,95%,and 100%) respectively.The size of plaque was continuously measured every day for 18 times with Logistic Model and Ratkowsky Modeland.At last,the conditions of toxin production by A.flavus were optimized by means of the Response Surface Model.The results showed that under the optimized culture conditions (35 ℃,100% humidity at a culture time of 16~17 days),the concentration of the extracted aflatoxin B 1 was 3 312 ng/mL,and the aflatoxin B 1 yield of per unit area was much higher than that (1 101.471 ng/mL) extrated before optimization in our lab.Further study is needed for carcinogenic mechanism of aflatoxin B.
