目的:探讨鬼针草治疗实验性干眼症的疗效和作用机制。方法:将18只成年新西兰白兔随机分为正常对照组、模型组、鬼针草治疗组,每组6只。模型组和鬼针草治疗组点用阿托品滴眼液建立干眼模型,鬼针草治疗组同时给予鬼针草灌胃,连续4周。实验前和实验后2周、4周对角膜荧光染色和泪液分泌量进行眼表评估,并在第4周取泪腺标本行免疫组织化学染色检测IL-1、TNF-琢、Fas和bcl-2表达。结果:模型组2周角膜荧光染色阳性,4周加重,鬼针草治疗组2周时角膜荧光染色阳性,但较模型组轻,且4周时未加重;泪液分泌量方面,模型组实验2周、4周较实验前明显下降(P<0.O5);鬼针草治疗组实验后SIT较实验前下降,但无统计学意义(P>0.05),与模型组相比,明显优于模型组(P<0.05);鬼针草治疗组泪腺腺泡上皮细胞中Fas和IL-1蛋白阳性表达量较模型组减少,差异有统计学意义(P<0.05)。结论:鬼针草能改善干眼兔的角膜上皮损伤和泪液分泌,能够抑制泪腺组织的凋亡和炎性反应,对干眼症泪腺组织具有保护作用。
Objective:To explore the effects and function mechanismof sticktight in treating experimental dry eye.Methods:All18mature white rabbits from New Zealand were randomly divided into the normal control group,the model group,the sticktight treatment group,each group six rabbits.The model group and the sticktight treatment group were given atropine eye drops to establish dry eye model,the sticktight treatment group was given intragastric administration of sticktight,continuous medication for four weeks.Two and four weeks before and after experiment,fluorescein staining(FS)was measured,tear secretion was evaluated through ocular surface.IL-1,TNF-,Fas andbcl-2expressions in lacrimal gland were detected by IHC in the fourth week.Results:FS in the model group of two weeks was positive,after four weeks,positivity got more high,FS in the sticktight treatment group of two weeks waspositive,but it was lower than the model group,positivity didn忆t also get more high after four weeks;tear secretionin the model experimental group of two and four weeks was dcreased obviously compared with before(P<0.05);SITin the sticktight treatment group was decrased obviously than before,but there was no statistical difference(P>0.05),compared with the model group,better than the model group obviously(P<0.05);Fas and IL-1protein expressions ofepithelial cells in lacrimal glands in the sticktight treatment group were decreased compared with the model group,there were significant statistical differences(P<0.05).Conclusion:Sticktight can improve corneal epithelium damage and tear secretion of rabbit with dry eye,inhibit apoptosis and inflammation reaction of lacrimal glands tissue,and has protection function for lacrimal glands tissue of dry eye.
