目的本实验拟阐明大鼠嗅上皮神经干细胞(NSCs)的免疫原性,为临床NSCs移植治疗和免疫排斥反应的防治提供一定的实验依据。方法本实验通过将成年大鼠嗅上皮分离、培养出巢蛋白免疫反应阳性并能分化为神经元和星形胶质细胞的NSCs。将大鼠嗅上皮NSCs分别与自体以及异体外周血单核细胞(PBMCs)共培养,共分为4组,使用cck-8比色法测定刺激后外周血单核细胞增殖能力,观察不同来源NSCs刺激PBMCs的增殖情况。结果异体的嗅上皮NSCs对于PBMCs的刺激增殖能力明显强于同体细胞。结论嗅上皮NSCs在体外有一定的免疫原性,可促进PBMCs增殖,移植时需注意免疫排斥问题。
Objective To explore the immunogenecity of neural stem cells (NSCs) from olfactory epithelium in vitro. Methods NSCs from zinc sulfate in situ injured adult rat olfactory epithelium were cultured in DMEM/F12 ( 1 : 1 ) containing 10% beat-in-activated fetal bovine serum. NSCs and specific neural cells differentiated from NSCs were identified with indirect immunohistochemistry. Different origin of NSCs was added into different experimental groups and the proliferation ability of peripheral blood mononuclear cells (PBMCs) was measured by CCK-8 methods. Results CCK-8 showed that there was no significant difference between group C and positive-control group (P 〉 0.05) ; there was significant difference between group B (D (450 nm) : 0.289±0.0275) and negative-control group and between group C (D (450 rim) : 0.476±0.0283) and negative-control group (P 〈0. 05). Conclusion NSCs from olfactory epithelium have immunogenecity in vitro and can stimulate the proliferation of PBMCs. Attention should be paid to inmmunologic rejection in the transplantation.
