[目的]探讨清热化湿方对幽门螺杆菌(helicobacter pylori,Hp)感染人胃癌SGC-7901细胞磷酸化PKC、CD14及MMP-1表达的影响。[方法]参照血清药理学方法制备清热化湿方含药血清,常规培养SGC-7901细胞株及Hp菌株(悉尼株,Sydney strain of Hp,SS1),取数对生长期人胃癌SGC-7901细胞,分别加入5%、10%、15%的清热化湿方含药血清预培养2h,再按细菌与细胞100∶1比例加入Hp培养6h,并以常规培养SGC-7901细胞为对照。应用实时荧光定量PCR技术、免疫印迹(Western-blot)检测Hp悉尼株感染对人胃癌细胞磷酸化PKC、CD14及MMP-1表达的影响及清热化湿方含药血清的调控作用。[结果]Hp感染人胃癌SGC-7901细胞后,磷酸化PKC、CD14及MMP-1表达较空白组明显增加(P〈0.05)。清热化湿方药物血清以剂量依赖的方式下调Hp感染胃癌SGC-7901细胞PKC磷酸化,并降低CD14及MMP-1表达。[结论]清热化湿方通过调控PKC磷酸化,下调Hp诱导的CD14及MMP-1表达,是其防治Hp感染相关胃癌的可能机制之一。
[Objective]To investigate the effects of Qingre Huashi decoction(QHD)on helicobacter pylori(Hp)infected human gastric cancer SGC-7901 cells in the regulation of the PKC phosphorylation and the expression of CD14 and MMP-1.[Methods]According to the method of serum pharmacology,the serum containing QHD was prepared.The SGC-7901 cell strain and Hp strain(Hp,SS1)were conventionally cultured.The logarithmic growth phase of huaman gastric cancer SGC-7901 cells were selected,added with serum containing QHD of 5%,10%,15% respectively and precultured for 2h.Then Hp was added according to the ratio of 100:1 between bacteria and SGC-7901 cells and cultured for 6h.The routine culture SGC-7901 cells used as control.Real-time PCR and Western blot were applied to test the expression of CD14 and MMP-1and the effect of serum containing QHD.[Results]After the SGC-7901 cells were infected by Hp,the expression of CD14 and MMP-1 as well as the PKC phosphorylation increased significantly compared with the control group(P〈0.05).The serum containing QHD downregulated the expression of CD14andMMP-1 as well as the PKC phosphorylation in a dose-dependent way.[Conclusion]QHD can downregulate the expression of CD14 and MMP-1 which were induced by Hp through the phosphorylation of PKC.It may be one of the possible mechanisms of the prevention and treatment of Hp infection in gastric cancer.
