目的研究不同浓度的青蒿素对人胰腺癌细胞JF305增殖的抑制作用,并初步探讨其作用机制。方法常规体外培养人胰腺癌JF305细胞,加入不同浓度的青蒿素处理48 h。倒置显微镜下观察细胞形态的改变,MTT法检测青蒿素对细胞增殖情况的影响,Hochest33258荧光染色法观察细胞凋亡形态学改变。结果倒置显微镜结果表明,随着药物浓度的增加,细胞数目逐渐减少。MTT结果表明,青蒿素对JF305细胞的增殖具有明显的抑制作用,且抑制作用随药物浓度的增加而增强。Hochest33258荧光染色结果表明,给药组细胞发生典型的凋亡形态改变。结论青蒿素对胰腺癌细胞JF305的增殖有明显的抑制作用,并能诱发细胞凋亡。
Objective To investigate the inhibitory effect of different concentration artemisinin on human pancreatic cancer cell line JF305 and its possible mechanism. Methods The human pancreatic cancer cell of JF305 were cultured in vitro and treated with different concentrations of artemisinin for 48 h. The change of cell morphology was observed under the inverted microscope. The proliferation of JF305 was measured by MTT assays. The changes of apoptotic morphologywere observed by method of Hochest33258 fluorescent staining. Results The inverted microscope observation result indicated that the number of cells decreased with the increase concentration of the artemisinin. And MTT results indicated that artemisinin has a significant inhibitory effect on the proliferation of JF305 cells in dose dependent manner. Hochest33258 fluorescence staining showed that the typical apoptotic morphological changes occurred in artemisinin treatment group. Conclusion Artemisinin could inhibit the proliferation of pancreatic cancer cell JF305 and induce apoptosis.
