目的:探讨涤痰汤对帕金森病大鼠多巴胺神经细胞凋亡及相关基因表达的影响。方法:应用6-羟基多巴胺立体定向注射制作PD大鼠模型。将成功的PD模型随机分为模型组与化痰组,另外纳入正常对照组和假手术组,分别给予相应处理,共14天。用TUNEL测定各组大鼠的细胞凋亡数,免疫组化染色法测定Bc l-2基因和Bax基因的表达。结果:与模型组比较,化痰组细胞凋亡数明显减少,差异有显著意义(P〈0.05);同时Bc l-2的表达明显增多,Bax的表达明显降低,差异均有显著意义(P〈0.01)。结论涤痰汤通过增强Bc l-2和降低Bax的表达,减少PD大鼠的神经细胞凋亡而发挥保护神经元的作用。
Objective:To study the effects of Ditan decoction on the apoptosis and related genetic expression of rates with Parkinson disease.Methods:The PD model rats were established through stereotoxic microinjection of 6-OHDA.The successful models of PD were randomized into the model group and the eliminated phlegm group,and we brought into the normal control group and the sham-operated group.Every group was given relevant treatment for 14 days.Then we counted the number of apoptosis of each group with TUNEL and determined the expression of Bcl-2 and Bax gene with immunohistochemistry.Results:Compared with the model group,the number of apoptosis of the eliminated phlegm group is significantly reduced,the difference is significance(P〈0.05),meanwhile the expression of Bcl-2 gene significantly increased and Bax gene significantly reduced,the difference is significance(P〈0.01).Conclusion:Ditan decoction could improve the expression of Bcl-2 gene and reduce the expression of Bax gene,then reduce the apoptosis of rats with PD to protect the nerve cells.
