目的:观察黄连解毒汤、黄连单煎剂对胰岛素抵抗大鼠氧化应激和内质网应激的影响。方法:采用高脂高热卡饮食饲养8周制备胰岛素抵抗大鼠模型,成模后随机分为黄连解毒汤纽(HJD组)、黄连单煎剂组(HD组)、Alpha-硫辛酸组(ALA组)和模型组(MOD组),各组以相应的药物干预4周。另设正常组(NOR组),予普通饲料喂养,不予药物干预。比较各组大鼠空腹血糖(FBG)、空腹胰岛素水平(Fins)和胰岛素敏感性(ISI)以观察药物对大鼠糖代谢的影响,血清及肝脏中丙二醛(MDA)、超氧化物歧化酶(SOD)、谷胱甘肽过氧化物酶(GSH—Px)活性以观察药物对大鼠氧化和抗氧化的影响。Western blot方法测定肝组织中的内质网应激标志物c—Jun氨基端激酶(c-jun NH2-terminal kinase,JNK)和Phospho—c-Jun NH2-terminal kinase(Ser73)(p-c—Jun)的蛋白含量变化,RT-PCR方法检测肝组织内质网应激标志物葡萄糖调节蛋白GRP78(glucose regulated protein78,GRP78)mRNA表达水平,以观察黄连解毒汤,黄连单煎剂对胰岛素抵抗大鼠内质网应激的影响。结果:与NOR组比较,MOD组大鼠FBG升高,Fins明显升高,ISI明显降低(P〈0.01),血清及肝脏中MDA含量明显升高(P〈0.05),SC)D、GSH—PX活性明显降低(P〈0.05,P〈0.01);与MOD组相比,HJD组,HD组大鼠FBG下降,Fins明显降低,ISI明显升高(P〈0.05,P〈0.01),SOD、GSH—Px的活性显著升高(P〈0.01,P〈0.05),MDA含量有一定程度下降但差异无显著性。比较各纽大鼠肝组织JNK蛋白表达水平,各组间差异无显著性;应激标志物pc-Jun/JNK的水平,MOD组较NOR组明显升高(P〈0.01),HJD、HD组较MOD组均明显下降(P均d〈0.01)。比较各组肝组织中内质网应激标志物GRP78/Bip mRNA水平,MOD组较NOR组显著升高(P〈0.05),HJD、HD组及ALA组较MOD?
OBJECTIVE To study the effects of Huanglian Jiedu decoction(HJD) and Huanglian decoction HD) on oxidative stress and endoplasmic reticulum stress of insulin resistant rats, and its possible molecular mechanism. METHODS Ninety male Wistar rats were fed with high-fat and high-caloric diet to establish insulin resistance model. After 8 weeks feeding, serum fasting blood glucose(FBO), fasting insulin(Fins), insulin sensitivity index(ISI) were determined. Fift-four insulin resistant rats were successfully induced and were randomly divided into model control group, the HJD group, the HD group and the alpha-lipoic acid group. The other 12 normal rats were used as control group. After administration with corresponding drugs for 4 weeks, the rats were sacrificed and serum FBG, Fins, ISI were determined, and the malonaldehyde(MDA), superoxide dis mutase(SOD), glutathione peroxidase (GSH-Px) in the serum and liver tissue were determined. The GRP78 mRNA expression in the liver was determined by reverse-transcription polymerase chain reaction (RT-PCR). Western blot analysis was employed to detect liver Phospho-c-Jun/JNK protein expression. RESULTS Compared with control group, model group showed higher FBG and Fins level while significantly decreased ISI level (P〈0. 01), higher MDA level while significantly decreased SOD and GSH-Px activity(P〈0. 05) in serum and liver tissue. In HJD, HD and ALA groups, FBG and Fins were decreased relative to model group, while ISI level were drastically increased as compared with model group(P〈0. 05, P〈0.01 ). The SOD and GSH-Px were elevated drastically relative to model group(P〈0. 01 or P〈0. 05), while MDA level were decreased as compared with model group.
