目的探讨参佛胃康汤剂对胃癌细胞的抗瘤作用及其机制。方法予昆明种小鼠皮下接种小鼠胃癌细胞MFC造模,将造模后的小鼠随机分为模型组和参佛胃康高、中、低剂量组;在肿瘤移植后的第1天ig给予实验组小鼠参佛胃康汤剂,在肿瘤移植后第61天处死各组小鼠。定期检测小鼠的生存状态、肿瘤大小及重量;采用TUNEL法和流式细胞仪测定肿瘤细胞的凋亡指数;采用免疫组化法检测肿瘤组织中血管内皮生长因子(VEGF)、STAT3、缺氧诱导因子-1αa(HIF-1α)的表达。结果与模型组比较,参佛胃康组小鼠肿瘤的大小和重量明显受到抑制,其凋亡指数显著增加;He瘤组织中VEGF、STAT3、HIF-1α的表达下调。结论参佛胃康汤剂可能通过诱导肿瘤细胞凋亡和下调VEGF、STAT3、HIF-1α等信号转导蛋白的表达来发挥其抗肿瘤作用。
OBJECTIVE To explore the anti - tumor effects of Shenfu Weikang decoction(SWD) on mice with gastric carcinoma and the mechansim. METHODS Kunming mice grafted with a mouse gastric adenocarcinoma cell line MFC was used as the experimental model. The mice were divided into control group, high, middle and low does of SWD groups. Mice in the three experimental groups received SWD over a 60 days period starting at the first day after grafting. All the mice were sacrificed at the 61th day after being graf- ted. The tumor size and weight were determined. Apoptotic indices were examined by the TUNEL method and the flow cytometric analy- sis. The expression of VEGF, STAT3 and HIF - 1 α in tumor tissues were examined and analyzed by immunostaining method. RESULTS Compared with control group, tumor size and weight were significantly inhibited, the Apoptotic indices in gastric cancer grafted mice were significantly increased, and the expression of VEGF, STAT3 and HIF - 1 α in the tumor tissues were down - regulated after treated with SWD. CONCLUSION The in vivo anti - tumor effect of SWD is related with the induction of the cell apoptosis and down - regulation of VEGF, STAT3 and HIF - 1 α which were the signal transduction pathway.
