目的研究雪灵芝水提取物对人胃腺癌MGC-803细胞Bcl-2和Bax基因表达的影响。方法不同浓度的雪灵芝水提取物处理体外培养的MGC-803细胞,采用四甲基偶氮噻唑蓝(MYr)法检测细胞存活水平;应用流式细胞术检测细胞周期和细胞凋亡水平;采用逆转录聚合酶链反应(RT—PCR)和蛋白印迹法(Western Blot)测定人胃腺癌MGC-803细胞的Bcl-2和Bax基因表达情况。结果雪灵芝水提取物作用于人胃腺癌MGC~803细胞后,0.1~0.8mg/mL浓度组细胞存活水平随药物浓度的增大而减小(P〈0.05);0.8mg/mL浓度组MGC-803细胞G.期比例为(61.23±6.45)%、S期细胞比例为(26.86±3.38)%、细胞凋亡率为(0.07±0,01)%,与对照组相比,P〈0.05;与对照组相比,0.8mg/mL浓度组Bcl-2 mRNA表达降低、Bax mRNA表达增加(P〈0.05);0.2~0.8mg/mL浓度组Bcl-2蛋白表达降低、Bax蛋白表达增加。结论雪灵芝水提取物在一定浓度范围内可降低MGC-803细胞的存活水平,使细胞发生凋亡,其机制可能为通过下调Bcl-2基因、上调Bax基因表达,促进细胞凋亡发生。
Objective To study the effect of Arenaria Kansuensis Aqueous Extract (AKAE) on the expressions of Bcl - 2 and Bax gene in human gastric cancer cell line MGC - 803. Methods MGC - 803 cells were incubated with AKAE at different concentration , cellular survival level was tested by MTT;Cell cycle and apoptosis were detected by flow cytometer (FCM) ; RT - PCR and Western Blot were used to detect the gene expression level of the Bcl - 2 and Bax. Results After treatment with AKAE, MGC - 803 cellular survival level gradually decreased at dose of 0.1 - 0.8mg/mL ( P 〈 0.05 ) ; The percentage of MGC - 803 G1 phase was (61.23 ± 6.45 ) %, S phase was (26.86 ± 3.38) % and the apoptotic level was (0.07 ± 0.01 ) % at dose of 0 . 8mg/mL, compared with control group( P 〈 0.05 ) ;At the dose of 0.8mg/mL, the Bcl- 2 mRNA expression decreased and the Bax mRNA expression increased, respectively compared with control group(P 〈 0. 05) ;At the dose of 0. 2 -0.8mg/mL, protein expression of Bcl -2 was decreased while protein expression of Bax was increased. Conclusion AKAE can decrease the Bcl -2 mRNA and protein expression and increase the Bax mRNA and protein expression in MGC -803 cells, so that it plays a role in inducing cell apoptosis.
