目的:研究复方蜥蜴散不同微粒组合剂对溃疡性结肠炎大鼠MMP-9和COX-2蛋白表达的影响,探讨其可能作用机制。方法:将84只SPF级雄性大鼠随机分为空白组,模型组,复方蜥蜴散80目组、100目组、(80+100)目组,柳氮磺胺吡啶组,除空白组外,其余组均采用2,4,6-三硝基苯磺酸-乙醇复合法诱导UC大鼠模型。造模成功后,各组大鼠分别做相应处理,光镜观察肠黏膜病理组织学变化,采用免疫组化法检测MMP-9和COX-2蛋白的表达。结果:(1)各治疗组MMP-9和COX-2蛋白的阳性表达率显著低于模型组(P〈0.01);(2)复方蜥蜴散(80+100)目组MMP-9和COX-2蛋白的阳性表达率显著低于复方蜥蜴散80目组、100目组和柳氮磺胺吡啶组(P〈0.05);(3)经Pearson相关分析,MMP-9和COX-2蛋白表达呈正相关(P〈0.05),相关系数r=0.969。结论:MMP-9和COX-2在UC过程中具有协同作用;复方蜥蜴散不同微粒组合剂可能通过降低MMP-9和COX-2的表达而抑制炎性细胞,减轻炎症反应,调节免疫,恢复肠道屏障,改善血液流变性和微循环,达到修复肠黏膜、促进溃疡愈合的作用。
Objective:To study different combinations of Compound Lizards Powder on expressions of MMP-9 and COX-2 in rats with ulcerative colitis and explore the underlying mechanisms. Methods:84 SPF male SD rats were randomly divided into blank group,model group,Compound Lizards Powder 80 mesh group,100 mesh group,(80+100)mesh group,sulfasalazine group,except blank group,other groups were collected 2,4,6 trinitrobenzene sulfonic acid-ethanol combined molding induced rat ulcerative colitis model. After the success of model,rats were treated accordingly,light microscope was used to observe pathological,expression of MMP-9 and COX-2 protein were detected by immunohistochemistry. Results:(1)The positive expression rates of MMP-9 and COX-2 protein in each treatment group was significantly lower than model group(P〈0.01).(2)The positive expression rate of MMP-9 and COX-2 protein in Compound Lizards Powder(80+100)group was significantly lower than compound lizards powder 80 mesh group,100 mesh group and sulfasalazine group(P〈0.05).(3)By Pearson correlation analysis,MMP-9 and COX-2 protein expression were positively correlated(P〈0.05),correlation coefficient r=0.969. Conclusion:MMP-9 and COX-2 have synergy in the UC process;Different combinations of Compound Lizards Powder may reduce the expression of MMP-9 and COX-2 to inhibit inflammatory cells,reduce the inflammatory response,regulate immunity,restore the intestinal barrier,improve blood rheology and microcirculation,to repair the intestinal mucosa and promote healing of ulcers.
