目的:探讨仙茅多糖(COP)对免疫低下小鼠免疫功能的调节作用。方法:从72只昆明种小鼠中随机留取12只作为正常对照组,剩余60只采用环磷酰胺腹腔注射建立免疫低下模型,并随机分为模型对照组,COP高、中、低剂量组和阳性对照组,每组12只。COP高、中、低剂量组分别给予400、200、100mg·kg-1·d-1浓度的COP溶液灌胃,阳性对照组给予200mg·kg-1·d-1浓度的香菇多糖溶液灌胃,正常对照组和模型对照组给予等体积0.9%氯化钠溶液灌胃,每日灌胃1次,共灌胃处理10d。给药结束后观察小鼠免疫器官指数,流式细胞术检测血细胞中T细胞亚群,观察NK细胞杀伤能力和小鼠脾脏T、B细胞转化能力,用酶联免疫吸附试验(ELISA)法检测小鼠血清中(TNF-α)含量。结果:与正常对照组比较,模型对照组小鼠的胸腺和脾脏指数下降、脾淋巴细胞转化能力显著降低(P〈0.05),仙茅多糖高、中剂量组和阳性对照组均可显著提高模型小鼠的脾脏和胸腺指数和脾淋巴细胞转化能力;模型对照组小鼠外周血中CD4+T亚群数量和CD4+T/CD8+T比值较正常对照组显著下降(P〈0.05),经COP治疗后CD4+T亚群数量和CD4+T/CD8+T比值均能得到有效恢复,尤以中剂量组效果最佳(P〈0.05);COP可以显著提高免疫低下模型小鼠血清中的TNF-α含量(P〈0.05)。结论:COP能明显改善环磷酰胺所致免疫低下小鼠的免疫功能。
Objective: To discuss the effect of Curculigo orchioides polysaccharide on the immunity of immunocompromised mice induced by cyclophosphamide. Methods: Twelve Kunming mice were selected randomly as the normal control group, and immunocompromised mice models were established in another 60 Kunming mice by intraperitoneal injection of cyclophosphamide. Then the mice models were randomly divided into 5 groups: model control group, positive control group, and Curculigo orchioides polysaccharide groups of low dosage, medium dosage, and high dosage. Mice in the treatment groups were given Curculigo orchioides polysaccharide solution of the concentrations at 100, 200, and 400mg·kg-1·d-1 respectively and those in the positive control group were given lentinan solution at 200mg·kg-1·d-1. The normal control group and model control group were given normal saline of the same amount. All groups received treatment by gavage once a day for 10 days. The immune organ indexes, the killing function of NK cells and the transformation ability of T-cells and B-cells in the spleen were observed, T-cell subgroups were detected with flow cytometry, and the TNF-α level in serum were measured with ELISA. Results: Compared withthe blank group, the thymus index, spleen index and transformation ability of splenic lymphocyte of the model group decreased significantly(P〈0.05). And these indexes were improved significantly in the high and middle dosage groups of Curculigo orchioides polysaccharide and the positive control group. The number of CD+4 T cells and the CD+4/CD+8 ratio in the peripheral blood were obviously smaller in the model group than those in the normal group(P〈0.05), which were restored effectively by Curculigo orchioides polysaccharide, especially at the medium dosage(P〈0.05). The level of TNF-αin the Curculigo orchioides polysaccharide group was also remarkably improved(P〈0.05).
