通过醛化法将膝沟藻毒素(GTX2,3)分别与血蓝蛋白(KLH)、牛血清白蛋白(BSA)偶联制备完全抗原,高效液相色谱法(HPLC)检测醛化反应前后游离GTX2,3的浓度,完全抗原免疫Balb/C小鼠,间接酶免疫吸附试验检测免疫后血清中抗体效价。GTX2,3与KLH及BSA偶联反应前溶液中毒素的浓度分别为760μg·mL^-1和660μg·mL^-1,偶联反应后毒素的浓度分别为191μg·mL^-1及256μg·mL^-1,Balb/C小鼠经过5次免疫后血清的效价为1:500。结果表明,反应后游离的GqN2,3的浓度降低,制备的GTX2,3完全抗原免疫小鼠后小鼠产生抗GTX2,3抗体,说明GqN2,3与载体蛋白发生偶联反应,偶联物具有免疫原性。

The immunogen of GTX2,3 were produced successfully through the methods of formaldehyde, and the synthesized GTX2,3 -KLH conjugates were improved for the antibody production of GTX2,3 as immunogen.
