目的:观察复方银黄微型灌肠剂(YHCE)对发热大鼠下丘脑中前列腺素E2受体EP1、EP4 mRNA表达的影响。方法:大鼠皮下注射20%酵母混悬液造发热模型,复方银黄微型灌肠剂按35.84g/ks于造模1h后灌肠给药;给药后4h取大鼠下丘脑组织,用实时荧光定量聚合酶链反应检测各组大鼠下丘脑中EP1、EP4受体mRNA表迭的相对差异。结果:与发热对照组比,YHCE组的EP1、EP4受体mRNA相对表达量减少。结论:复方银黄微型灌肠剂可抑制发热组大鼠下丘脑中前列腺素E受体EP1、EP4 mRNA的表达。
Objective: To determine the effect of yinhuang microenema(YHCE) on the expression of prostaglandin E2 receptors EPI and EP4 mR- NA in the febrile rat hypothalamus. Method: 24 male Wistar rats were divided randomly into 3 groups: normal control group( n = 8), fever group( n = 8), and YHCE group( n = 8 ). Except the control group, all rats were injected 20% suspension of yeast hypodermically ( 10ml/ kg). One hour later, the YHCE were administered . Then we taken out the hypothalamus tissue of the rat at 4 hours after administered and stored at - 80℃. Examined the expression of prostaglandin EP1 and EP4 receptors mRNA by the real - time fluorescence quantitative polymerase chain reaction. Result: Compared with fever group,the expression of prostaglandin EP1 and EP4 receptors mRNA in the YHCE group were markedly lower (P 〈0.05). Conclusion: YHCE inhibit the expression of EP1 and EP4 mRNA in febrile rats hypothalamus.
