为检测日本血吸虫小RNA let^#(Sja-let^#)对虫体生殖发育的影响,本研究在日本血吸虫尾蚴感染BALB/c小鼠后的11 d^#2 d,将Sja-let^#的同系物(Sja-let^# mimics)注射入宿主体内对血吸虫Sja-let^#的表达进行干扰,结果显示Sja-let^# mimics干扰组小鼠肝脏虫卵孵化率显著升高;小鼠肝脏肉芽肿病变减轻十分明显,病灶周围炎性细胞浸润半径变小,空泡变性较对照组轻微,周围正常组织完整性也优于对照组。扫描电镜和透射电镜未观察到虫体的明显变化。利用荧光定量PCR检测Sja-let^#的候选靶基因干扰虫体的表达情况,结果表明Sja-let^# mimics对大部分候选基因存在不同程度的抑制作用。本研究为后续深入研究小RNA Sja-let^#的功能奠定了基础。
To study the fimction of Sja-1et-7 on reproductive development of Schistosoma japonicum, the expression of Sja-1et-7 was interfered by injected Sja-1et-7 miRNA mimics into the mice during 11 to 42 days after inoculating S.japonicumvia the tail. The results shown egg hatching rates in Sja-1et-7 mimics group were significantly increased. The liver granulomatous lesions of Sja-1et-7 mimics group were relieved obviously, the surrounding inflammatory cells and vacuoles degeneration were mild, the surrounding normal tissue was better than that in control group. No significant changes on the body structures were observed under scanning electron microscopy and transmission electron microscopy. The expression of Sja-1et-7's candidate target genes, the expressions of most related genes were suppressed by Sja-1et-7 miRNA mimics to a certain degree detected by real-time PCR. This study might provide some foundation for the further study on the function of Sja-1et-7.
