目的 确定羊抗鼠IgG胶体金分子的构象。方法 应用原子力显微镜扫描生理状态下分布在云母表面的羊抗鼠IgG胶体金分子,并应用随机附带的软件进行统计分析。结果 羊抗鼠IgG胶体金分子为22.0nm×9.0nm×5.7nm的“椭圆球状”结构。结论 羊抗鼠IgG胶体金分子具有均一稳定的特征性构象。原子力显微镜可以用于测定生物大分子纳米尺度的结构。
Objective To identify the configuration of goat anti- mouse IgG gold conjugate (IgG- gold). Methods The molecules of IgG - gold affiliated on the new cleaved mica were scanned with atomic force microscope (AFM) under the physiological environment. The data was analyzed with the statistic software in the AFM. Results The configuration of IgG - gold molecule is an "ellipsoidal sphere" sized as 22.0 nm× 9.0nm× 5.7 nm. Conclusion IgG - gold has a steady uniform characteristic conformation. AFM can play an important role in detecting biomacromolecules at a nanometer scale resolution.
