非重型再生障碍性贫血患者血清对32D细胞凋亡及Stat3蛋白磷酸化的影响

目的 :研究鹿龙再生汤对重型再生障碍性贫血(severe aplastic anemia,SAA)小鼠模型外周血象及红系爆式集落形成单位(burst-forming unit-erythroid,BFU-E)和粒单系集落形成单位(colony-forming unit-granulocyte macrophage,CFU-GM)的影响,探讨鹿龙再生汤促进造血作用的机制。方法 :BALB/c小鼠40只通过干扰素(interferon,IFN-γ)腹腔注射联合白消安灌胃,连续用药10 d,成功诱导建立了SAA模型,然后被随机分为4组(n=10),分别设为SAA组、SAA中药小剂量组(一般浓度)、SAA中药中剂量组(正常剂量的5倍浓度)、SAA中药大剂量组(正常剂量的10倍浓度),并分别予以生理盐水及3个不同剂量的鹿龙再生汤灌胃;另取10只正常BALB/c小鼠作为正常组,予以生理盐水灌胃和腹腔注射。5 d后收集各组小鼠眼眶血检测外周血象,取小鼠骨髓单个核细胞用甲基纤维素培养,培养7 d后观察并计数的BFU-E、CFU-GM。结果 :造模后SAA小鼠外周血的白细胞、血红蛋白、血小板、网织红细胞均有降低(P〈0.05),经过不同剂量的鹿龙再生汤治疗后SAA小鼠的外周血均有不同程度升高,其中中剂量的鹿龙再生汤可以明显升高SAA模型小鼠外周血白细胞和网织红细胞的计数(P〈0.05)。SAA组BFU-E和CFU-GM的集落形成较正常组明显下降(P〈0.05),而SAA中药中剂量组BFU-E和CFU-GM的集落形成较SAA组明显上升(P〈0.05)。结论 :中剂量的鹿龙再生汤可能通过促进SAA小鼠骨髓单个核细胞BFU-E、CFU-GM的生成,从而改善外周血象,相关实验结果可能为SAA的中药治疗提供理论依据。

英文摘要:

Objective: To study the effects of Lulongzaisheng soup on burst-forming unit-erythroid(BFU-E) and colony- forming unit-granulocyte macrophage(CFU-GM) in severe aplastic anemia(SAA) mouse model and explore its mechanisms of promoting the hematopoiesis. Methods: 40 BALB/c Mice were randomly divided to 4 groups(n=10, respectively) which were selected to set up the SAA model using intraperitoneal injection with IFN-γ and intragastric administration with busulphan. These mice were named as the low-dose, middle-dose and high-dose groups according to the different dose soups used, which were equaling to the same concentration in human begings, 5 times of the concentration and 10 times of the concentration, respectively. Another 10 BALB/c mice were selected to intragastric administration with the same volume sterilized distilled water. After 5 days, the orbital venous blood was taken to assay the blood routine. Bone marrow mononuclear cells were collected and cultured in methycellulose 7 days, then counting the number of BFU-E, CFU-GM. Results: Compared with the normal group, the number of peripheral blood cells in the SAA group was decreased(P〈0.05, respectively). After treated by different dose Lulongzaisheng soup, the number of peripheral blood cells was increased, and the middle-dose Lulongzaisheng soup can improve the leukocytes, the absolute reticulocytes count and the number of BFU-E CFU-GM(P〈0.05). Conclusion: The middle-dose Lulongzaisheng soup may increase the number of peripheral blood cells by improving the count of BFU-E, CFU-GM of Bone marrow mononuclear cells in SAA mice. These results may provide the theoretical basis for traditional Chinese medicine treatment of aplastic anemia.
