目的:探讨中药复方菩人丹(PRD)对OLETF大鼠视网膜中自杀相关因子(fas)和半胱天冬酶-8(caspase-8)表达的干预作用。方法:将24只成模的OLETF大鼠随机分为PRD治疗组、糖尿病模型组,每组均为12只,同周龄12只雄性LETO大鼠为正常对照组,PRD治疗组大鼠连续灌胃PRD,1.8g/kg,1次/d,2个月。HE染色观察大鼠视网膜的形态结构改变;TUNEL法检测OLETF大鼠视网膜神经细胞的凋亡情况;免疫组化法和Western Blot方法检测OLETF大鼠视网膜fas和caspase-8表达的蛋白;RT-PCR方法检测各组大鼠视网膜中fas和caspase-8 mRNA的表达情况。结果:与正常对照组LETO大鼠相比,OLETF模型组的大鼠视网膜出现了明显的病理变化,神经细胞的凋亡指数、fas、caspase-8的蛋白及mRNA表达明显升高(P〈0.01)。与模型组大鼠相比,PRD治疗组大鼠视网膜的病理变化明显减轻,神经细胞的凋亡指数、fas、caspase-8蛋白及mRNA表达明显降低(P〈0.01)。结论:PRD可通过抑制fas、caspase-8的表达,减少糖尿病大鼠视网膜神经细胞的凋亡,从而起到保护糖尿病视网膜神经组织的作用。
Objective: To investigate the interventions of traditional chinese medicine PRD on the factors associated suicide (fas) and cysteine aspartic acid specific protease-8 (caspase-8)in retina of the OLETF rats. Methods:24 rats were randomly divided into two groups, a PRD treatment group and a diabetes model group and 12 male LETO rats of same age as a normal control group, PRD 1.8 g/kg (l/d) was intragastrically administrated to the rats of PRD treatment group for 2 months. Then rats retinal morphology were observed by Hematoxylin-eosin staining and apoptosis of OLETF rat retinal neurons were detected by TUNEL. Besides, immunohistochemical staining and Western Blot were used to detect caspase-8 and fas in protein level. RT-PCR was used to detect caspase-8 and fas in mRNA level. Results:Compared with normal control LETO group, the neuronal apoptosis index, the protein and mRNA expression of fas and caspase-8 of dia- betes model OLETF group were significantly increased ( P 〈 0.01 ), but compared with the model group, neuronal apoptosis index, the protein and mRNA expression of fas and caspase-8 protein in the PRD treatment group were significantly decreased (P 〈 0.01 ). Conclusion:PRD can inhibit fas and caspase-8 expression, reducing neuronal apoptosis in diabetic rat retina, which play a protective role in diabetic retinal nerve tissue.
