检测新城疫病毒(NDV)感染和poly(I∶C)刺激后A549细胞的Toll样受体3(TLR3)、LC3-II水平变化,并在TLR3过表达以及干扰表达后检测LC3-II、p62的表达水平,以探讨TLR3与NDV诱导细胞自噬之间的关系。结果表明:poly(I∶C)刺激后激活TLR3通路,能够引起细胞自噬;过表达TLR3后,LC3-Ⅱ的表达在NDV感染6h后高于对照组,在感染后期p62降解程度明显;干扰TLR3表达后,NDV诱导LC3-Ⅱ的表达水平受到抑制,在感染后期p62降解程度减弱。证明TLR3参与NDV诱导的细胞自噬作用。
The roles of Toll-like receptor 3(TLR3)in Newcastle disease virus(NDV)-induced autophagy of A549 cells were explored in this study.The expression of TLR3 and LC3-II was detected by Western-blot after NDV infection or poly(I∶C)stimulation.In TLR3-overexpressing or TLR3-knockdown cells after NDV infection,the conversion of LC3-Ιto LC3-II and expression of p62 were analyzed by Western-blot.The result demonstrated that expression of TLR3 and LC3-II gradually increased with the NDV infection or poly(I∶C)stimulation.In TLR3-overexpressing cells,NDV led to higher expression of LC3-Ⅱand stronger degradation of p62 while in TLR3-knockdown cells,the expression of LC3-Ⅱwas slightly inhibited and degradation of p62 was slowed down.The results indicated that NDV could activate TLR3 and its signal pathway,which may have an effect on autophagic level caused by NDV.
