高原世居健康藏族和汉族人外周血线粒体I、II、III、IV和V复合物相关编码基因mRNA表达量比较

目的比较高原世居健康藏族和汉族人外周血线粒体I、II、III、IV和V复合物相关编码基因mRNA表达量,初步了解藏族人线粒体复合物编码基因mRNA的表达差异。方法本研究采用实时定量PCR方法,检测青海玉树地区健康藏族男性和汉族男性外周静脉血线粒体复合物I、II、III、IV、V编码基因NDUFA1、SDHA、UQCRC1、COX6A2、ATP6VOD2mRNA表达量,并行比较研究。结果发现复合体IV中的基因COX6A2和复合体V中的ATP6VOD2基因的表达量之间存在差异,藏族人COX6A2和ATP6VOD2基因表达量均高于汉族人,而其他编码基因无差异。结论藏族人线粒体复合物编码基因mRNA的表达差异主要集中在线粒体电子传递链的细胞色素氧化酶复合体IV的COX6A2和复合体V的ATP6VOD两个基因。

英文摘要:

Objective To compare and detect the expression levels of mRNA in the mitochondrial electron transfer chain complex ( I - V) coding genes NDUFA1, SDHA, UQCRC1, COX6A2, ATP6VOD2 in healthy native high altitude dwellers - Tibetans and Chinese Hans. Methods 6 healthy Tibetans and Chinese Hans were recruited. Their blood was collected into BD PAXgene RNA tubes and store in fridge and the total RNA were isolated and ex- tracted according to PAXgene RNA kit. The expressions of mitochondrial complex I, II, III, IV, V coding genes were detected with florescent quantitative real - time PCR. Results The mRNAs expressions of complex IV ( COX6A2), and complex V(ATP6VOD2) in Tibetans were significantly up - regulated compared with Han. Conclusion The mR- NAs expression levels in mitochondrial complex IV(COX6A2) and complex V(ATP6VOD2) coding genes might be the possible reasons for Tibetans well adapted in Qinghai - Tibetan Plateau, and which would be a basic research data for future studies of molecular adaptation mechanisms in Tibetans.
