目的建立一种丹参淀粉含量检测方法 ,测定不同产地样品淀粉含量,分析淀粉与有效成分含量间的关系。方法采用高氯酸-蒽酮比色法测定丹参植物干燥根中淀粉含量,进行方法学考察,并测定山东、河南、四川3个产地20株丹参样品,同时采用超高效液相色谱(UPLC)方法对样品中8种有效成分进行测定,分析二者的相关性。结果建立了一种丹参植物干燥根淀粉含量测定方法。该方法在0~50 mg/L内具良好的线性关系,具有良好的精密度、重复性和加样回收率,RSD均小于3%,方法准确可靠;不同产地丹参样品中淀粉的含量与丹参酮类成分呈正相关趋势,与丹酚酸类成分呈负相关趋势。结论丹参根中淀粉的积累与有效成分的代谢密切相关。
Objective To establish a method for determination of starch content in Salvia Miltiorrhiza(SM), detect starch contents of samples from different producing areas by the method and analyze the relationship between starch and effective constituents. Methods After a systematic study on methodology, a perchloric acid- anthranone colorimetric method was introduced to detect starch contents of 20 samples from Shandong, He′ nan and Sichuan. An ultra performance liquid chromatography(UPLC) method was used to detect effective constituents contents in samples. The relevance between them was researched. Results A determination method for starch content was established which had a good linearity(0-50 mg/L), excellent accuracy, repeatability and recovery(RSD〈3%). A negative correlation trend of starch contents was found with liposoluble constituents contents and a positive correlation trend was found with hydrosoluble constituents contents in SMs from different origins. Conclusion It reminds a close relationship between starch accumulation and effective constituents metabolism in SMs.
