酞酸二丁酯(DBP)具有一定的内分泌干扰作用,对环境健康产生不利作用.采用DBP抗体,建立了间接竞争性时间分辨荧光免疫分析方法(TRFIA),测定了镇江市内水域中DBP含量,以风险熵法评估其生态风险.结果显示:优化后的TRFIA方法半数抑制率IC50值为35.8 ng·mL~(-1),最低检测限为2.4 ng·mL~(-1),检测线性范围为4.7~272.6 ng·mL~(-1).该方法特异性高,DBP抗体与7种DBP结构类似物的交叉反应率低于4.65%,加标回收率为79.85%~119.20%,批内差为5.54%~9.93%,批间差为7.27%~14.01%.镇江市内河水中的DBP检出率为100%,质量浓度为5.89~94.57 ng·mL~(-1),河水中DBP分布存在较高的生态风险.
As an endocrine disruptor,dibutyl phthalate( DBP) has adverse effects on environment and human health. Using a polyclonal antibody of DBP,a sensitive indirectly competitive time-resolved fluorescence immunoassay( TRFIA) was proposed, and several parameters were investigated. DBP concentration in the rivers of Zhenjiang was determined,and the ecological risk was assessed by risk quotients. The results show that under the optimized conditions,the 50% inhibition concentration( IC50)value of TRFIA is 35. 8 ng·mL~(-1)with limit detection value of 2. 4 ng·mL~(-1)in the detection range from4. 7 to 272. 6 ng·mL~(-1). The developed method shows high selectivity with cross-reactivities less than4. 65% to seven structurally related phthalate esters. The recoveries are from 79. 85% to 119. 20%,and the reproducibilities of intra-assay CV are from 5. 54% to 9. 93% with reproducibilities of inter-assay CV from 7. 27% to 14. 01%. The detection rate of DBP in the rivers of Zhenjiang city is 100%,and the concentration ranges from 5. 89 to 94. 57 ng·mL~(-1). There exists high ecological risk for DBP distribution in rivers.
