番茄斑萎病毒属(Tospovirus)是布尼亚科(Bunyaviridae)唯一能够侵染植物的病毒属。本研究于2012—2013年对云南省昆明市的园林绿化景区和蔬菜种植区进行调查,采集13份疑似番茄斑萎病毒属病毒感染的植物样品,并通过特异性引物进行RT-PCR扩增,克隆和测序,获得完整的核壳体蛋白基因(nucleocapsid protein,NP)全序列,结合Genbank数据库中的数据,构建番茄斑萎病毒属属病毒NP蛋白的系统发育树。结果表明:13个样本中有11个样本被番茄斑萎病毒属病毒侵染,包括番茄斑萎病毒、马蹄莲褪绿斑病毒、番茄环斑病毒、凤仙花坏死斑病毒、朱顶红褪绿环斑病毒;寄主包括蜘蛛兰、蝴蝶兰、番茄和尖尾芋、曼陀罗、旱金莲、鸢尾、裂叶喜林芋,其中尖尾芋是番茄斑萎病毒属病毒入侵的新寄主。
Tospovirus is the only genus to infect plants in the Bunyaviridae family. During 2012--2013, 13 samples that possibly infected by Tospovirus were collected from scenic landscaping and vegetable-growing field in Ktmming, Yunnan Province. The complete nucleic acid sequence of nucleocapsid protein (NP) was obtained by using RT-PCR, cloning and sequencing. The phylogenetic tree was constructed using MEGA 5. 05 software based on the amino sequence of NP. The result showed that 11 of 13 samples were identified as Tospovirus, including Tomato spotted wilt virus (TSWV), Calla lily chlorotic spot virus (CCSV), Toma- to zonate spot virus (TZSV), Impatiens necrotic spot virus (INSV) and Hippeastrum chlorotic ringspot vims (HCRV). The natural hosts are Hymenocallis littoralis, Phalaenopsis amabilis, Lycopersicon esculentum, Alocasia culcullata, Datura stramoniumL , Tropaeolum majus, Iris tectorumand Philodendron andreanum. A. culcullata is a new host of compound infection of Tospovirus.
