本文用GeNorm与NormFinder算法分析actin、α-tubulin、cblp和yptc1四个常用内参基因在不同处理下莱茵衣藻中的表达。结果显示,α-tubulin在各种条件下都有较好的表达稳定性,而以不同基因组合构成的多内参系统可能更有利于细胞周期短暂的单细胞绿藻的分析。
The selection of reference genes plays a key role in the study of gene expression by real-time quantitative PCR. Using GeNorm and NormFinder algorithms, we studied the expression of four common-used reference genes, actin, a-tubulin, cblp and yptcl, in Chlamydomonas reinhardtii under different treatments. Our results showed that the expression of a-tubulin was relatively stable, but it might be more reliable to use a combination of different genes and utilize their normalization factor (NF) as a reference for analyzing the dynamic changing gene expression of unicellular green algae.
