目的检测感染SIV初期用病毒巨噬细胞炎症蛋白-Ⅱ(vMIP—Ⅱ)治疗的食蟹猴外周血T细胞TCRVβ优势表达,分析特异性Vβ亚家族表达水平的变化及T细胞克隆性质的改变。方法11只食蟹猴感染前后外周血PBMC样本共22例通过半定量RT-PCR技术分析其中14个TCRVβ亚家族T细胞的表达情况,运用基因扫描技术分析用药组与感染组差异表达的T细胞TCRVβ亚家族基因中CDR3的长度了解其克隆性。结果与感染前正常食蟹猴外周血T细胞的14个TCRVβ亚家族中表达(除去表达量极低的10个亚家族)相比较,感染后SIV组中部分样品Vβ7、8、14、16的表达量与感染前比较有上升趋势;vMIP-Ⅱ组中TCRVβ亚家族的表达量上升更加明显。基因扫描结果显示感染前、后标本中Vβ7亚家族的表达由多克隆向寡克隆变化的趋势。结论体内实验研究表明,SIV感染初期的食蟹猴体内部分TCRVβ亚家族有特异性的增生,且克隆性发生改变。vMIP—Ⅱ对此种Vβ亚家族的表达的增生有促进的作用,推测其促使特异性应答的免疫细胞的增生。
Objective To explore the T cell receptor β chain variable region (TCRVβ) repertoire perturbations and T cell clonal expansion during primary SIV-infection and after treated with vMIP- Ⅱ in cynomolgus mokeys ( Macaca Fascicularis ). Methods The TCRVβ repertoire was analyzed by semi-quantitative polymerase chain reaction using PBMC samples obtained from 11 cynomolgus monkeys at preinfeetion and 17 days after infection. Then high sensitive gene scan technology, was used to identify the clonality of specific Vβ subfamily which demonstrated an expansion for SIV. Results Vβ7, 8, 14, 16 subfamilies were expanded in some cynomolgus monkeys in group of SIV and vMIP- Ⅱ . The gene scan analysis showed the quality of T cell expansion in dominant Vβ7 expression changed from polyclonal to oligoclonal in 12 sampies. Conclusion Some specific Vβ families are expanded during primary SIV-infection, and vMIP- Ⅱ may enhance their expression, which means that the therapy of vMIP- Ⅱ during primary infection mainly induce the expansion of immune cells and strengthen the function of immune system.
