毛细管电泳(Capillary Electrophoresis,CE)在痕量物质的分析与检测、样品的半制备与制备、多维分析以及联用技术中仍面临巨大挑战,研究者从降低电场强度、使用低电导率缓冲液、加入有机试剂、引入制冷,发展了毛细管内径大于150μm的大通道毛细管电泳技术(Wide-bore Eectrol phoresis,WBE)。本文综述了这一新型大通道毛细管电泳及其相关的多维和联用技术,展望了WBE的发展趋势及潜在应用。
Capillary electrophoresis (CE) has been widely applied in pharmaceutical, biological, food, chemical,environmental, and clinical fields and so on. However, CE is also facing great challenges in determination and analysis of trace samples, semi-preparation and preparation of samples, multi- dimensions analysis and coupling of multiple technologies. Low sample loading ability is one of the most important factors that affects the development of CE. It is thought that the direct increase of inner diameter of separation channel is an effective solution, but the increase of inner diameter of separation channel also induces the problem of Joule heat. Researchers have developed the wide-bore electrophoresis (inner diameter~ 150 /~m) technology by decreasing electric field intensity, using low conductivity buffer, adding organic reagent and introducing cooling mechanism and so on. This paper reviews the progress in wide-bore electrophoresis(WBE) and its related multi-dimension electrophoresis technologies and coupling technologies. Meanwhile, the development trend and potential applications of WBE technology are prospected.
