目的探讨热休克转录因子1(HSF1)对肿瘤坏死因子α(TNF-α)的转录调控作用及机制。方法构建HSF1真核表达载体pcDNA3.1-HSF1,TNF-α野生型启动子荧光素酶报告基因pGL3-TNFα-Y,TNF-α突变型启动子荧光素酶报告基因pGL3-TNFα-T。pcDNA3.1-HSF1转染巨噬细胞RAW264.7,半定量RT-PCR检测TNF-αmRNA的表达。pcDNA3.1-HSF1,TNF-α启动子荧光素酶报告基因转染RAW264.7,比较pGL3-TNFα-Y和pGL3-TNFα-T的相对萤光素酶活性。结果 HSF1下调RAW264.7 TNF-αmRNA的表达,pGL3-TNFα-T的相对荧光素酶值较pGL3-TNFα-Y明显下降,差异有统计学意义(P〈0.01)。结论热休克转录因子1(HSF1)通过与肿瘤坏死因子α(TNF-α)启动子区HSE的结合下调RAW264.7 TNF-αmRNA的表达。
Objective To study the transcriptional regulation of heat shock transcription factor 1(HSF1) on TNF-α and explore its mechanisms.Methods The eukaryotic expression vector containing mouse HSF1,pcDNA3.1-HSF1,luciferase reporter gene containing mouse TNF-α wild type promoter,pGL3-TNFα-Y,and luciferase reporter gene containing mouse TNF-α mutant type promoter,pGL3-TNFα-T,were constructed.The pcDNA3.1-HSF1 was transfected into mouse macrophage RAW264.7,and the expression of TNF-α mRNA was determined by semi-quantitative RT-PCR.The pcDNA3.1-HSF1 and the luciferase reporter gene containing mouse TNF-α promoter were co-transfected into RAW264.7,and the relative luciferase activities of pGL3-TNFα-Y and pGL3-TNFα-T were determined and compared.Results HSF1 could downregulate the expression of TNF-α mRNA in RAW264.7.The relative luciferase activity of pGL3-TNFα-T was significantly lower than that of pGL3-TNFα-Y(P〈0.01).Conclusions HSF1 downregulates the expression of TNF-α mRNA in RAW264.7 by binding with the HSE in the promoter of TNF-α.
