以七元瓜环为主体,抗癌药物甲氨蝶呤为客体,利用^1H NMR技术和紫外可见吸收光谱法研究了七元瓜环(Q[7])对甲氨蝶呤(MTX)的分子识别作用,同时利用相溶解度及溶出度法考察了Q[7]对MTX溶解度及溶出度的影响。结果表明:MTX与Q[7]可形成2:1的主客体复合物,平衡常数为(9.55±0.37)×10^11L^2.Q[7]能增加MTX的溶解度及溶出度,并且Q[7]对MTX的增溶效果受体系的酸碱性影响。最后用^1H NMR法进一步证实了主客体的作用模式。
In this paper, the interactions of cucurbit[ 7 ] uril( Q [ 7 ] ) with anti-cancer drug MTX in different proton forms were investigated in details by UV absorption spectroscopy, phase solubility studies and ^1H NMR technique. The effects of pH on complex were also explored. The result showed that the inclusion ratio of Q [7] and MTX was 2: 1, and the formation constant was (9.55 ±0.37) × 10^11 L^2.mol-2. Moreover, solubility and dissolution of methotrexate increased as a function of Q[7] concentrations. The inclusion complex was further proved by ^1H NMR.
