将体外培养的小鼠囊胚分别施以40℃1、2、3h和38℃1、2、3h热应激处理,然后在37℃条件下分别恢复3、2、1h后,提取胚胎细胞总RNA,用RT—PCR方法检测小鼠囊胚Hsp70基因的表达,以明确小鼠囊胚经热应激处理后是否有诱导型Hsp70基因表达。结果表明:各试验组,包括对照组鼠的囊胚都有诱导型Hsp70基因的表达。说明小鼠囊胚对热应激处理敏感,轻微热应激就可使诱导型Hsp70基因表达。
The mice blastocysts cultured in vitro were heat treated at 40 ℃ and 38 ℃ for 1, 2, and 3 h, respectively, and then were recovered at 37 ℃ for 3, 2 and 1 h, respectively, in order to detect the expression of Hsp70 gene in heat stress mice blastocysts. The total RNA in the embryonic cells was extracted and the expression of Hsp70 gene was detected by RT-PCR. The results indicated that each group of blastocysts, including the control group, had the expression of Hsp70 gene. And the above results indicated that the mice blastocysts are sensitivity to heat stress, therefore, a slight heat stress could induce Hsp70 gene expression.
