为探讨白介素10(IL-10)及其受体IL-10R相互作用并调节妊娠耐受的可能机制,在孕E4.5、E6.5和E8.5给BALB/c×C57BL/6孕鼠腹腔注射抗IL-10R抗体,而给NOD×C57BL/6孕鼠腹腔注射重组小鼠IL-10。在E10.5检测小鼠蜕膜NKG2D+TGF-β-NK细胞构成比和胚胎吸收率。研究发现,注射抗小鼠IL-10R抗体可显著增高BALB/c×C57BL/6孕鼠蜕膜NKG2D+TGF-β-NK细胞构成比,并相应地显著增高胚胎吸收率。相反,注射外源性IL-10可显著降低NOD×C57BL/6孕鼠蜕膜NKG2D+TGF-β-NK细胞百分率,达到接近于BALB/c×C57BL/6孕鼠生理状态下同源细胞的水平,并可显著降低NOD×C57BL/6小鼠胚胎吸收率。这些结果提示,IL-10和IL-10R相互作用可调节小鼠蜕膜NKG2D+TGF-β-NK细胞百分率,并使其保持在较低水平,而这种状态有利于同种异基因妊娠成功。
To investigate the possible mechanism of interaction of IL-10 and IL-10 receptor(IL-10R) and modulation of feto-maternal tolerance,multiple injection of the anti-IL-10R antibody was given intraperitoneally into pregnant BALB/c×C57BL/6 mice and multiple administration of recombinant mouse IL-10 was performed intraperitoneally into pregnant NOD×C57BL/6 mice at embryonic days E4.5,E6.5,E8.5.The proportion of decidual NKG2D+TGF-β-NK cells and the percentage of embryo resorption were determined on embryonic day E10.5.It was found that anti-IL-10R antibody administration significantly increased the proportion of decidual NKG2D+TGF-β-NK cells and the percentage of embryo resorption was significantly increased accordingly.In contrast,exogenous IL-10 treatment significantly decreased the percentage of decidual NKG2D+TGF-β-NK cells in NOD×C57BL/6 mice to a level comparable to that in BALB/c×C57BL/6 mice,and decreased the percentage of embryo resorption.These results suggest that IL-10 and IL-10R interaction may modulate the proportion of decidual NKG2D+TGF-β-NK cells and maintain it at a low level,and this status seems to be beneficial for the success of allogeneic pregnancy.
