目的探讨D-δ-三烯生育酚抗人乳腺癌MCF-7细胞的生物学活性。方法将细胞分为对照组(无药物培养)和实验组,实验组设低、中、高剂量组,分别加入终浓度为30,40,50μmol·L^-1的D-8-三烯生育酚,药物干预MCF-7细胞24h后,显微镜下观察各组细胞生长状态,四甲基偶氮唑盐(MTT)法、流式细胞仪分析细胞增殖、周期与凋亡。结果D-8-三烯生育酚抑制乳腺癌MCF-7细胞增殖,低、中、高剂量组细胞增殖抑制率分别为(9.33±1.68)%,(32.47±3.18)%和(76.63±1.00)%,与对照组比较,差异有统计学意义(P〈0.01);低、中、高剂量组G。期细胞所占比例分别为(54.34±1.50)%,(58.24±0.81)%和(62.11±1.12)%,与对照组[(43.34±2.51)%]比较,差异有统计学意义(P〈0.05);低、中、高剂量组贴壁细胞明显减少,漂浮死亡细胞明显增多,细胞早期凋亡率Q4分别为(10.87±0.50)%,(17.87±2.40)%和(25.83±3.76)%,与对照组[(1.93±0.25)%]比较,差异有统计学意义(P〈0.05)。结论D-8-三烯生育酚能抑制MCF-7细胞增殖,阻滞细胞于G1期,诱导细胞凋亡,具有抗乳腺癌MCF-7细胞的生物学活性。
Objective To explore the biological activity of D-5- tocotrienols against human breast cancer MCF-7 cells. Methods The Breast cancer MCF-7 cell line was treated in vitro with O(control), S0 (low),40 (middle), and 50 (high) μmol . L^-1 D-5-tocotrienols,respectively for 24 h. Cell growth was observed under the microscope. MTI" analysis and flow cytometry were applied to measure cell proliferation,cell cycle and apoptosis. Results D-8-tocotrienols inhibited the proliferation of MCF- 7 cell line. The cell proliferation inhibition rates of the low, middle, and high dose groups were(9.33± 1.68 )%, (32.47±3.18)% and(76.63±1.00) %, respectively, compared with the control group. The differences were statistically significant (P〈0.01). The percentage of G1 phase cells was(54.34±1.50) %, (58.24±0.81) % and(62.11±1.12) % ,respectively in the experimental groups, all significantly higher than ( 43.34 ±2.51 ) % in the control group ( P 〈 0.05 ). The percentage of adherent cells was significantly reduced, and the floating dead cells significantly increased in the experimental groups ,The rates of early apoptosis were significantly increased by ( 10.87 ±0.50) %, ( 17.87±2.40) %, and (25.83 ±3.76 ) %, respectively, compared to that by ( 1.93± 0.25 ) % in the control group (P〈0.05). Conclusion D-5-tocotrienols has anti-breast cancer activity by inhibiting the proliferation of MCF-7 cell line,leading to cell cycle arrest at G1 phase and inducing apoptosis.
