体外分离培养成人脂肪组织来源的间充质干细胞,探索其基本生物学特性及向心肌样细胞诱导分化的潜能。体外分离培养人脂肪干细胞并诱导其向脂肪、成骨和软骨细胞分化,采用倒置相差显微镜观察细胞的形态学特点,用油红、碱性磷酸酶、von Kossa和甲苯胺蓝染色鉴定其分化情况,流式细胞仪测定细胞表面抗原CD34、CD44、CD45及HLA-DR的表达,采用血管紧张素Ⅱ诱导人脂肪干细胞向心肌样细胞分化,并与5-氮胞苷诱导结果对比。结果表明:分离培养的脂肪干细胞呈"梭形"生长,核大;流式细胞仪检测阳性表达CD44,CD105,阴性表达CD34,CD45及HLA-DR;脂肪、成骨和软骨的诱导分化证明所分离培养的细胞是间充质干细胞。血管紧张素Ⅱ诱导4周后,心肌肌钙蛋白cTn-I、缝隙连接蛋白43、肌球蛋白重链MHC呈阳性表达,但未见细胞的自发性搏动。脂肪干细胞具有间充质干细胞的特征,在一定条件下能向心肌样细胞分化,是一种有潜力的治疗心肌梗死的细胞源;血管紧张素Ⅱ能诱导人脂肪干细胞向心肌样细胞分化,可以用来替代传统的诱导剂5-氮胞苷。
The basic biological characteristics of adult adipose-derived stem cells (ADSCs),which were isolated and cultured from adult adipose tissue in vitro,and the potential of differentiating into cardiomyocytes by chemical induction were studied in this paper.ADSCs were isolated and cultured in vitro,and were induced into adipocyte,chondrocyte and osteoblast.The morphological characteristics of cells were observed by the phase-contrast microscope,and the ability of differentiation was observed by Oil Red,alkaline phosphatase,von Kossa and Toluidine Blue staining.The expressions of CD34,CD44,CD45,CD105 and HLA-DR were detected by flow cytometry.The ADSCs were induced with angiotensin Ⅱ to differentiate into cardiomyocyte-like cells,and compared with the 5-azacytidine.The isolated and cultured ADSCs presented fusiform shape,and ADSCs expressed CD44,CD105 but not CD34 CD45 and HLA-DR by flow cytometry.The differentiations to adipocyte,chondrocyte and osteoblast proved the cells with the characteristics of stem cells.After 4-week induction by angiotensin Ⅱ,the cells expressed myosin heavy chain,troponin-I and connecxin43 by immunocytochemistry staining,but without beating of the cells.ADSCs have the characteristics of mesenchymal stem cells and angiotensin Ⅱcan induce ADSCs to cardiomyocyte-like cells.
