本研究利用qRT-PCR技术检测了绵羊卵巢周期性黄体变化过程中Ghrelin/MC4R mRNA水平相对表达变化规律。结果表明随着黄体的形成、发育、退化,成熟黄体Ghrelin mRNA表达量显著高于新鲜黄体(P〈0.05),极显著高于退化黄体(P〈0.01)。而MC4R基因呈现了相反的趋势,退化黄体MC4R mRNA表达量极极显著高于新鲜黄体和成熟黄体(P〈0.001)。Ghrelin/MC4R在绵羊卵巢周期性黄体相左的表达模式,揭示了这两种基因参与黄体周期性变化,且与生殖密切关系。
We used the qRT- PCR to detect the expression of ghrelin/MC4R in sheep ovary throughout the estrous cycle. In fact, the relative mRNA levels of ghrelin varied depending on the stage of tile cycle, with the highest expression during the development of die corpora lutea (CL) and minimal expression in the regressing CL(P 〈 0.01 ). But MCAR showed the reversed pattern, relative mR- NA levels of MC4R varied depending on the stage of the cycle, with the highest expression levels in regressing CL and lowest expression at the young and mature phases ( P 〈0. 001 ). Dynamic changes of in the profile of ghrelin expression during the estrous cycle suggest a precise regulation of ovarian expression of ghrelin/MCAR, which could represent a potential role for them in the regulation of luteal development.
